Multiplex Assay Overview:
The assay service provided by Eve Technologies on their Human Cytokine Array / Chemokine Array 71-Plex facilitates research in any area that involves the immune system such as inflammation, allergies, irritable bowel disease (IBD), sepsis, and cancer. This Discovery Assay® measures forty-eight biomarkers simultaneously in a single microwell and twenty-three biomarkers simultaneously in another microwell. The assay is designed for extracellular protein analysis in nearly all types of biological samples.
“Cytokine” is a general term used for a diverse group of soluble proteins and peptides which act as regulators under both normal and pathological conditions to modulate the functional activities of individual cells and tissues. These proteins also mediate direct interactions between cells and regulate processes taking place in the extracellular environment. Cytokines differ from hormones in that they act on a wider spectrum of target cells. Also, unlike hormones, they are not produced by specialized cells which are organized in specialized glands. The cytokine group of proteins includes lymphokines, interferons, colony stimulating factors and chemokines. Cytokine and chemokine research plays a significant role in achieving a deeper understanding of the immune system and its multi-faceted response to most antigens, as well as disease states such as inflammatory disease, allergic reactions, irritable bowel disease (IBD), sepsis, and cancer.
Intended Sample Type:
Human serum, plasma (EDTA recommended), cell cultures, tissue homogenates, synovial fluid, CSF, sputum, BAL, urine and others. Due to a wide range of sample conditions, a pilot run is recommended for cell cultures, tissue homogenates, and other sample types not listed.
Special Sample Preparation:
ALL samples must be centrifuged prior to aliquoting. The aliquot should be drawn from the middle of the sample volume. Samples that have a high amount of contaminants, cellular debris, lipids, or coagulation can result in a volume loss as volume is being displaced by this debris and can clog the filters. Highly turbid or coagulated samples that clog the filters will be partially filtered by piercing the clogged membrane.

Luminex xMAP technology is used for multiplexed quantification of 71 Human cytokines, chemokines, and growth factors.
The multiplexing analysis is performed using the Luminex™ 200 system (Luminex, Austin, TX, USA) by Eve Technologies Corp. (Calgary, Alberta). Seventy-one markers are simultaneously measured in the samples using Eve Technologies’ Human Cytokine 71-Plex Discovery Assay® which consists of two separate kits; one 48-plex and one 23-plex (MilliporeSigma, Burlington, Massachusetts,USA) according to the manufacturer’s protocol. The 48-plex consists of sCD40L, EGF, Eotaxin, FGF-2, FLT-3 Ligand, Fractalkine, G-CSF, GM-CSF, GROα, IFN-α2, IFN-γ, IL-1α, IL-1β, IL-1RA, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IL-10, IL-12 (p40), IL-12(p70), IL-13, IL-15, IL-17A, IL-17E/IL-25, IL-17F, IL-18, IL-22, IL-27, IP-10, MCP-1, MCP-3, M-CSF, MDC, MIG/CXCL9, MIP-1α, MIP-1β, PDGF-AA, PDGF-AB/BB, RANTES, TGFα, TNF-α, TNF-β, and VEGF-A. The 23-plex consists of 6CKine, BCA-1, CTACK, ENA-78, Eotaxin-2, Eotaxin-3, I-309, IL-16, IL-20, IL-21, IL-23, IL-28A, IL-33, LIF, MCP-2, MCP-4, MIP-1δ, SCF, SDF-1α+β, TARC, TPO, TRAIL, and TSLP. Assay sensitivities of these markers range from 0.14 – 55.8 pg/mL for the 71-plex. *Note: %CV is an appropriate measure of quality only for samples that have significant detectable concentrations for a particular analyte. Low concentration/low signal samples can often have a much higher %CV as the signals are at the limitations of the assay and fall within a flat portion of the curve.
